The altercation began shortly before 3 p.m. inside Marriott's Residence Inn on Sixth Avenue in Midtown.
Police said a man approached the reception desk and demanded a room.
When hotel staff refused, the man became belligerent and was asked to leave.
A security guard was escorting the man out of the hotel when the man punched him once in the chest and ran, jumping into a cab.
The 44-year-old security guard collapsed unconscious and was rushed to Bellevue Hospital Center, where he was listed in critical condition.
Police said it was unclear what caused the security guard to collapse.
The suspect was described as a male in his 20s.
A woman who answered the phone at the Residence Inn yesterday declined to comment and said no one was available to speak about the incident.
